Description

Objectives

(A) Provide advanced knowledge on the techniques most used in the separation, structural elucidation, characterization and quantification of organic compounds. (B) Provide the fundamental concepts of the various analytical and structural methods, which qualitative and quantitative information may be obtained, and which are the advantages and disadvantages / limitations. (C) Provide an integrative view of the complementarity and applicability of the various analytical and structural methods. (D) Choose an analytical techniques depending on the sample. (E) Determine the concentration of the element or compound in the sample (and associated error). (F) Apply the acquired knowledge to practical cases. (G) Promote the autonomy of students in finding solutions to proposed problems. (H) Improve the skills of teamwork.

Syllabus

Atomic spectroscopies (elementary): emission in UV-Vis (Inductively Coupled Plasma - Atomic Emission Spectrometry(ICP-OES) and mass detection (ICP-MS)). Molecular spectroscopies: UV-Vis (absorption, fluorescence and phosphorescence), infrared (IV), Raman and circular dichroism. Fourier transform infrared spectroscopy (FTIR). Nuclear Magnetic Resonance (NMR) of Solids and Liquids (2D and 3D). Electronic paramagnetic resonance spectroscopy (EPS). X-ray crystallography. Separative Methods: Gas Chromatography; High Performance Liquid Chromatography. Supercritical Fluid Chromatography. Capillary electrophoresis. Mass Spectrometry (MS): Atomic (Isotopes; GC-C- IRMS); Molecular (MALDI-TOF-MS; ESI-MS). Hyphenated Methods (GC x GC-ToF-MS) Proteomics (Two-dimensional gel electrophoresis (2DE)) and metabolomics.

Prerequisites

Electromagnetic character of light and interaction with matter; Chemical bond; Periodic properties of the elements; Chemical Equilibrium: acid-base, solubility and complexation; Simple concepts of electrochemistry; Functional groups and nomenclature; Errors and statistics: associated error and other parameters, simple linear regression.

Cross Competence Component

In the curricular unit of MAE I, the main components of transversal skills will be developed, including oral expression, written expression, teamwork, critical and innovative thinking, and problem solving strategies. It should be noted that emphasis will be placed on continuous assessment in the TP and L components.

Laboratorial Component

Lab classes are mandatory. A strong connection will be maintained between the theoretical and laboratory components, with lab work based on material previously covered in the theoretical classes. Students will be organized into groups of 3 elements.

Programming And Computing Component

-----

Ethical Principles

All members of a group are responsible for the group´s work. In any assessment, every student shall honestly disclose any help received and sources used. In an oral assessment, every student shall be able to present and answer questions about the entire assignment and solution.
